What is Tauopathy?
Tauopathy is a class of neurodegenerative diseases brought about by the abnormal clumping of Tau protein in the human brain. This aggregation occurs when there is hyperphosphorylation of the protein, which eventually leads to unstable microtubules. Tauopathy diseases include Alzheimer’s Disease, Progressive Supranuclear Palsy, and Primary Age-related Tauopathy.
Tauopathies are closely related to but should not be confused with synucleinopathies. The latter are neurodegenerative diseases that occur as a result of the abnormal aggregation of the alpha-synuclein protein. Synucleinopathies include Parkinson’s disease, multiple system atrophy (MSA), and dementia with Lewy bodies (DLB). Interaction between the Tau and alpha-synuclein proteins may lead to the overlapping of these conditions.
What are the Common Symptoms of Tauopathies?
Symptoms of tauopathy include:
- Changes in Personality – Deposits of abnormal tau proteins in the frontal lobe of the brain brings about behavioral and personality changes. This may include withdrawal from social situations. Patients can also become highly irritable, lose motivation, and become increasingly moody.
- Inconsistent Sleep Patterns – Changes in the hypothalamus, the part of the brain that enables transition into sleep, results in changes in sleep patterns. Nightmares and insomnia may be common.
Like many other neurodegenerative diseases, tauopathy interferes with the transmission of messages between nerve cells inside the brain. That affects multiple areas related to cognitive, physical, and organ function in the body.
One of the approaches used by scientists to treat tauopathies is eliminating hyperphosphorylated Tau. Manipulation of molecular chaperones whose function is to refold misfolded proteins is one method through which defective Tau can be removed. Studies have shown that Hsp90, a type of molecular chaperone, is capable of degrading tangled Tau in mice and cell cultures.
Tau Protein Pre-Formed Fibrils
Naturally, Tau fibrils are self-assembled aggregates produced through fibrillation of the protein at high temperatures. The Tau is then refolded into a misfolded state. These fibrils are, therefore, useful in research involving the accumulation of defective tau proteins in brain neurons. Studies have shown that injection of pre-formed fibrils can result in Tau aggregates similar to neurofibrillary tangles.
